The Effect of Certain Inhibitors and Acti- Vators on Sperm Metabolism*

چکیده
The semen used in these studies was collected from healthy dairy bulls by means of an artificial vagina and was kept in a refrigerator until used. All experiments were begun within 2 hours after the semen was collected. Observations were made on the effect of inhibitors on bull sperm in the egg yolk-buffer pabulum previously described (2, 3). Semen and yolk-buffer at pH 6.8 were mixed at the ratio of 1:3 and stored at 10”. Motility and lactic acid production were studied in a modified Ringer-phosphate medium of the following composition: 0.9 per cent NaCl ,100 parts, 1.15 per cent KC1 4 parts, 2.11 per cent KHzPOd 1 part, 0.25 M Na2HP04 20 parts. Enough 1 N HCl was added to bring the mixture to pH 6.8. When this medium was used, the spermatozoa were removed from the seminal fluid by centrifuging and washed by suspending in 0.9 per cent NaCl. After centrifuging again the spermatozoa

Background: For understanding the roles of fatty acids on the induction of acrosome reaction which occurs under association of cholesterol efflux and PKA or PKC pathways in boar spermatozoa, metabolic fate of alone and combined radiolabeled 14C-oleic acid and 3H-linoleic acid incorporated in the sperm was compared, and behavior of cholesterol and effects of PKA and PKC inhibitors upon fatty aci...

Background: Mammalian spermatozoa are highly susceptible to lipid peroxidation, which leads to structural damage to the sperm cell, the enzymatic leakage from spermatozoa accompanied by lowered motility and metabolism. Catalase, an enzyme that detoxify hydrogen peroxide and reduce the enzyme leakage from sperm cell Materials and Methods: For this study, buffalo bull testicles (20 pairs) were pi...
متن کاملIn Vitro Inhibition of Human Sperm Creatine Kinase by Nicotine, Cotinine and Cadmium, as a Mechanism in Smoker Men Infertility
Background Nicotine, cotinine and cadmium are harmful components of cigarettes that have an effect on human reproductive function. Although the effects of cigarette smoke on male reproductive function is characterized in several articles its mechanism of action is still unknown. In the present study, we investigate the effect of nicotine, cotinine and cadmium on human sperm creatine kinase acti...
متن کاملI-16: Computer Aided Sperm Analysis andSperm Functional Testing (Hyperactivation) asBackground Tools in the Evaluation of SpermFunction/Quality
After twenty years Computer Aided Sperm Analysis has developed considerably and is now routinely used in many laboratories across the world. The advantage of the CASA methodology available is twofold: Analysis of particularly sperm concentration, sperm motility, sperm morphology and vitality is quantified in an Background manner. Secondly, most of the CASA analysis with the exception of sperm m...
متن کاملInhibition of Sperm Respiration and Reversibility of the Effects of Metabolic Inhibitors*
During the course of an investigation of the effect of certain metabolic inhibitors on the fertilizing ability of spermatozoa it became necessary to know whether or not the effects of the inhibitors were reversible. The studies reported herein were undertaken to gain this information. In earlier studies (l-3) of the reversibility of inhibition certain agents were added to counteract the effect ...
